Is it possible to make a manuscript amendment once a document has been signed by all parties or will it need to be resent?

As you mentioned in your question, you need to resend it. 

 

After an envelope get completed (all signers have signed it) you cannot edit to add an amendment for example.

 

In cases like that, you need to copy that completed envelope, copying an envelope will bring the signed document, so you need to add the amendment document then send to the same recipients again to sign it.
